Abstract:
In der Hypodermis des Hummers findet sich ein Ferment, das β-Glucoside des N-Acetyl-d-glucosamins unter Bildung von N-Acetyl-d-glucosamin spaltet. N-Acetyl-β-d-glucosaminidase aus Aspergillus oryzae wurde frei von Lactase erhalten. Sie hydrolysiert Methyl-, äthyl-, n-Propyl-, Benzyl- und Phenyl-N-acetyl-β-d-glucosaminid sowie N.N'-Diacetyl-chitobiose und Lacto-N-biose II, nicht aber Lacto-N-tetraose und Lacto-N-biose I. Das pH-Optimum liegt bei pH4, die Michaelis-Menten-Konstante für äthyl-N-acetyl-β-d-glucosaminid als Substrat beträgt ˜ 0.005 Mol/Liter. Aus N-Acetyl-glucosamin + äthanol erhält man durch enzymatische Synthese β-äthyl-N-acetyl-glucosaminid frei von der α-Verbindung.
