 Abstract: CO2 dark fixation is a basic metabolic process, related with the tricarboxylic acid (TCA)
cycle and biosynthetic processes, starting from the pool of TCA cycle intermediates. In bacteria
phospho-enolpyruvate-carboxylase (PEP-carboxylase) is one of the key enzymes for carboxylation of C3 -compounds under formation of oxaloacetate. The fine regulation of PEP-carboxylase is rather complex. TCA cycle intermediates can be inhibitors. The CO2 dark uptake reflects
the metabolic activity of the bacterial cell: only in the presence of degradable substrates 14-CO2
is taken up. The rate of ta CO2 uptake depends on the amount of available substrates. Growth
estimates derived from 14-CO2 -uptake and independent direct growth estimates with a digitizer-
microcomputer system on scanning electron micrographs agree rather well.
