 Zusammenfassung: Publisher Summary: This chapter examines mechanisms involved in small intestinal transport of calcium and phosphate ions. In an experiment described in the chapter, brush border membranes (BBM) were isolated by an Mg++-precipitation method from rabbit duodenum. Basal–lateral plasma membranes (BLM) were isolated by a Percoll centrifugation method from rat upper small intestine. Transport studies were carried out by a rapid filtration technique. When Ca-transport was studied, membrane samples were washed in 1 ml of ice-cold buffer solution containing 1 mmol/l EGTA prior to filtration to remove Ca bound to the outside of membrane vesicles. All incubations were carried out at 25°C and pH 7.4. It was concluded that for Pi, the Na–Pi-cotransport system in the brush border membrane is a site of coupling of Pi-transport to the energy of the Na-gradient and a site of regulation of Pi-transport by 1,25-D3. Mechanisms for active transport of Ca are localized in the BLM: (1) an ATP-driven system and (2) a Na/Ca exchange system. Regulation of Ca permeability by 1,25-D3 can be demonstrated at the BBM.
