 Abstract: Fast and efficient syntheses of 2′,3′-dideoxynucleoside-5′-O-(α-thio)triphosphates (α-S-ddNTP's) starting from 2′-deoxynucleosides are described. They have been used in a modified DNA-sequencing method and were tested as potential inhibitors of HIV-1 reverse transcriptase. An X-ray structure of 3′-deoxythymidine is presented and compared to other known 2′,3′-dideoxynucleoside structures.
