 Abstract: Means for changing the filling solution of patch pipettes are highly desirable whenever the influence of solution composition on cellular processes is to be studied. This refers both to patch configurations (e.g. studies of ionic selectivity or drug action on the single channel level) and to tight-seal whole cell recording. In the latter case it is particularly interesting to load cells with second messengers, regulating proteins, antibodies or toxins. Such substances very often interfere with seal formation, demanding seal formation to be performed while the pipette tip contains inert solution. A fast exchange of pipette filling solution at later times allows a timed application of the substance of interest.
