Abstract: (from the Introduction:) In this chapter we give a brief overview of the application of fluorescence in quantitative cell biological studies, with special emphasis on Förster-type fluorescence resonance energy transfer (FRET) and intracellular trafficking, and address the advantages and potential pitfalls of different detection and labeling strategies from the standpoint of a cell or molecular biologist.
